主要研究方向

转录是细胞特异性解读基因组的第一步,也是决定细胞功能和命运的关键步骤。本实验室利用荧光成像,基因组学,生物化学和基因组编辑多种研究手段,研究细胞分化模型和癌症细胞模型中在染色质环境下转录调控的分子机理,以及开发人为控制基因表达的新方法。活细胞中的分子事件是动态发生的而非静止不变的:转录因子在细胞核内与DNA的结合是高度动态和瞬时的,RNA也以爆发式的形式产生。本实验室开发和利用活细胞高速单分子荧光成像技术,研究转录调控因子与染色质的相互作用和在细胞核中的单分子动态,从而阐释转录因子控制细胞状态和命运的机理。染色质在三维空间的构象对远程转录调控至关重要。通过合成特异转录因子可以实现改变染色质三维构象和基因的转录活性。本实验室开发荧光标记基因组DNA的方法,研究染色质空间构象调控的关键蛋白因子,从而探索转录在三维空间的调控机理。
