摘要:Analyses of gene expression across large numbers of single cells can reveal cell-to-cell variability. To this end, my lab has been developing single-cell RNA-seq methods (e.g. Smart-seq2) that combine full-length transcript coverage with high sensitivity and accuracy. We have performed global analyses of gene expression in hundreds of individual cells from mouse preimplantation embryos and somatic cells. Using genetic crosses (CAST/EiJ x C57BL/6J), we leverage strain-specific SNPs to investigate X-chromosome inactivation, imprinting and allelic expression patterns of autosomal genes. Importantly, the analyses revealed striking allelic expression fluctuations that generated widespread random monoallelic expression in single cells in both stem cells and differentiated cells. Through single-cell transcriptome analyses of clonally related somatic cells, we distinguished between random monoallelic expression that was clonally propagated through cell division and dynamic fluctuations that arise as a consequence of stochastic transcription. Altogether, our results reveal a dynamic landscape of allelic transcription that is apparent in single cells and we discuss its functional consequences.
