摘要:Today, many life science applications depend on high data content assays and demand ever-increasing data quantity and quality.  To meet this demand, the throughput of assay readout technologies like microarrays, QPCR, sequencing, and imaging has advanced dramatically. Next-generation sequencing (NGS) is an impressive example where readout no longer limits throughput for small projects.  Three trends point to sample preparation as an increasingly restrictive bottleneck in future NGS workflows:  continuing reductions in readout cost, increasingly complex sample preparation for more advanced assays, and demand for higher sample throughput in research and clinical applications.  My lab seeks to debottleneck NGS, imaging, and screening workflows using advanced microscopy-compatible microfluidic sample preparation.  I will describe our work to integrate complete NGS sample preparation workflows using a lab-on-a-chip approach, further integrate sample generation in the same device, and establish other novel microfluidic platforms to address the sample preparation challenge.
