Abstract
 
Although detailed molecular mechanisms have been studied intensively in many areas of immunology, the immune repertoire – the sequence makeup of antibodies and T cell receptors (TCR) and the composition of B or T cells that express these receptors – has not yet been comprehensively characterized due to many technical limitations. In this talk, I will first discuss the development of a high-throughput sequencing based method to profile the antibody repertoire and its application in studying the antibody repertoire development. Next, I will discuss the development of a microfluidics based single cell gene expression analysis and its application in studying T cell thymic selection in combination with peptide-MHC tetramer staining. Then, I will talk about a force-based micropipette adhesion assay and its application in studying the mechanism of T cell receptor antigen recognition. Finally, I will outlook how we can better understand the immune system, modulate it, and personalize medicine and treatment using a systems approach.
