讲者介绍:Dr. Chiarle is a Professor in Pathology at Harvard Medical School. He received an M.D. degree from the University of Turin and did his postdoc training about B and T cell lymphomas with Giorgio Inghirami and Michele Pagano at New York University. He was a Visiting Professor in Fred Alt lab at Harvard Medical School, where he worked on chromosomal translocations and contributed to develop a new high-throughput method to clone translocations genome-wide. In 2012, he was appointed as Associate Professor of Pathology at Harvard Medical School and his lab is interested in mechanisms and therapy of cancers.  Dr. Chiarle has made significant contributions to elucidate the mechanisms that drive transformation of lymphocytes and his work has been published on Nature, Cell, Nature Medicine, Cancer Cell, Nature Cancer, et al.


报告摘要:Chromosomal translocations are major drivers of oncogenesis in hematological malignancies and solid tumors, yet the molecular mechanisms underlying their formation remain incompletely understood. This talk will discuss emerging mechanisms responsible for generating recurrent oncogenic rearrangements, with a particular focus on the interplay between DNA damage, genomic architecture, and endogenous sources of genome instability. We will examine the contribution of APOBEC-mediated mutagenesis and retrotransposable elements to DNA damage and chromosomal rearrangements and explore how these processes shape cancer genomes. ALK rearrangements in lymphomas and solid tumors will provide a paradigm for understanding how distinct mechanisms of genomic instability generate recurrent, tumor-promoting chromosomal translocations across different cellular and disease contexts.
